A computerized search for the appearance of heritable mutations (as indicated by changes in protein expression) was conducted on three sets of mice, whose sires had been either untreated, exposed to 3 gray units of gamma radiation, or treated with 150 mg/kg ethylnitrosourea. Proteins from the livers of approximately 800 mice were separated by two-dimensional electrophoresis, and abundances were measured by using image analysis techniques. Heritable mutations were detected by the appearance of new proteins or by the quantitative decrease in abundance of normally occurring proteins. Measurements of the variability of the protein abundance indicate that at least 48 proteins are consistent enough to be used in searches when mutations are expected to result in a 50% reduction in the normal amount of protein. New proteins were found in four offspring from ethylnitrosourea-treated mice, and in each case a nearby spot was found to be significantly diminished. These mutations were confirmed in subsequent generations. A computer-assisted search detected three of these mutations on the basis of the abundance decrease alone. These results indicate that two-dimensional electrophoresis can be used to detect mutations reflected as quantitative changes in protein expression, provided that the proteins to be monitored are quantitatively stable when samples from different individuals are compared. 相似文献

Control programmes against Echinococcus granulosus in its dog-sheep transmission cycle (Fig. 1) have been successful in many parts of the world'. In contrast, the related E. multilocularis presents a much more complex problem for control authorities. Unlike E. granulosus, the life cycle of E. multilocularis predominantly involves sylvatic hosts (e.g. rodents and foxes) (Fig. 2) and the control of wild life echinococcosis presents a formidable challenge to ecologists and epidemiologists. This review contrasts the two parasites, explaining why E. granulosus in its domestic dog-sheep life cycle has been so responsive to control, and examines the prospects for control of E. multilocularis. 相似文献

The development of the mesonephros and metanephros was examined in the marsupial Northern native cat, Dasyurus hallucatus, from birth through to the end of lactation. The mesonephros was present at birth, reached a maximum volume 11 days after birth and had regressed completely by day 30. The metanephros was present on day 2; glomeruli were first seen on day 8, and nephrogenesis continued until day 89 post partum. The newborn native cat is at a very primitive stage of development compared to other marsupials. Translocation of individuals among extant populations is an important tool in species conservation that allows managers to
supplement dwindling populations and potentially alleviate the deleterious effects of inbreeding. Ideal translocation strategy
should consider historical relationships among existing populations to avoid potential disruption of population subdivision
and local adaptation. Here, we examine mitochondrial sequence variation in the endangered blue duck Hymenolaimus malacorhynchos, a New Zealand endemic riverine specialist, to facilitate informed decision making in future translocations. Behavioural
observations suggest that blue duck dispersal is limited and may result in genetic structure within and between regional populations.
We analysed 894 base pairs of mitochondrial control region in 78 adult blue ducks sampled from 11 river catchments across
the species’ range (representing four regions in the North Island and three regions in the South Island) and found strong
and significant genetic structure both within and among islands. These results, combined with a 2.0% sequence divergence between
islands, indicates that North Island and South Island blue ducks should be treated as separate management units. The relationship
between genetic differentiation and geographic distance for blue ducks on the South Island conformed to an “isolation by distance”
pattern. Overall, we recommend that translocations of blue ducks should not be made between the North and the South Islands

Background
The objective of this study was to evaluate angiogenesis according to CD34 antigen expression in estrogen receptor (ER)-positive and negative breast carcinomas.Methods
This study comprised 64 cases of infiltrating ductal carcinoma in postmenopausal women divided into two groups: Group A: ER-positive, n = 35; and Group B: ER-negative, n = 29. The anti-CD34 monoclonal antibody was used as a marker for endothelial cells. Microvessel count was carried out in 10 fields per slide using a 40× objective lens (magnification 400×). Statistical analysis of the data was performed using Student's t-test (p < 0.05).Results
The mean number of vessels stained with the anti-CD34 antibody in the estrogen receptor-positive and negative tumors was 23.51 ± 1.15 and 40.24 ± 0.42, respectively. The number of microvessels was significantly greater in the estrogen receptor-negative tumors (p < 0.001).Conclusion
ER-negative tumors have significantly greater CD34 antigen expression compared to ER-positive tumors.57.
Human-induced changes to natural systems can cause major disturbances to fundamental ecological and population processes and result in local extinctions and secondary contacts between formerly isolated species. Extensive fur seal harvesting during the nineteenth century on Macquarie Island (subantarctic) resulted in extinction of the original population. Recolonization by three species has been slow and complex, characterized by the establishment of breeding groups of Antarctic and subantarctic fur seals (Arctocephalus gazella and Arctocephalus tropicalis) and presumed nonbreeding (itinerant) male New Zealand fur seals (Arctocephalus forsteri). One thousand and seven pups from eight annual cohorts (1992-2003) were analysed using mitochondrial control region data (RFLP) and 10 microsatellite loci to estimate species composition and hybridization. Antarctic fur seals predominated, but hybridization occurred between all three species (17-30% of all pups). Involvement of New Zealand fur seals was unexpected as females are absent and males are not observed to hold territories during the breeding season. The proportion of hybrids in the population has fallen over time, apparently owing to substantial influxes of pure Antarctic and subantarctic individuals and non-random mating. Over 50% of New Zealand hybrids and 43% of Antarctic-subantarctic hybrids were not F(1), which indicates some degree of hybrid reproductive success, and this may be underestimated: simulations showed that hybrids become virtually undetectable by the third generation of backcrossing. While human impacts seem to have driven novel hybridization in this population, the present 'time slices' analysis suggests some biological resistance to complete homogenization. 相似文献

Coniothyrium minitans, a mycoparasite of sclerotia of Sclerotinia sclerotiorum and Sclerotium cepivorum, produced four closely related metabolites inhibitory to fungal growth. The major metabolite, identified as macrosphelide A, had IG(50) values (the concentration of metabolite to inhibit growth by 50%) of 46.6 and 2.9 microgram ml(-1) against S. sclerotiorum and S. cepivorum, respectively. Fostering confers obvious advantages to the offspring but is seemingly costly to the caregiver. Such behaviour is particularly paradoxical in seals where the energetic investment in milk is very high and has led to the suggestion that this behaviour may have evolved through either kin selection or reciprocity. We used a combination of genetic and behavioural data to investigate whether kin selection plays a role in the fostering behaviour observed in a well-studied population of Antarctic fur seals (Arctocephalus gazella) from Bird Island, South Georgia. Genotypic data from eight highly polymorphic microsatellite markers were used to estimate relatedness among mother-pup pairs, foster mother-pup pairs and the total population. Mean relatedness was found to be significantly higher for foster mother-pup pairs than that observed for the total population, suggesting that kin selection could have a role in the maintenance of fostering behaviour in this species. 相似文献
